That wasn't a trans joke, it was a joke about the fake Ashley Madison profiles, and the fact that a lot of the "beautiful lady" profiles on those sites are actually men with fake accounts. The end.

The joke in question makes a joke out of the idea of men dressing in women's clothing. Transgender is an umbrella term that covers all kinds of people including cross dressers. Cross dressers are men that dress in women's clothing. There isn't really a joke there, it's just the idea of cross dressers that is supposed to be funny. I'm not a cross dresser, but I am trans and I get kind of tired of trans people being the butt of jokes.
