Albert Hofmann ("inventor" of LSD) was looking at amides of lysergic acid. The 25th was the diethyl, hence the nickname "25." He ingested some accidentally, probably through his skin, and began to trip.
Later, after he came down, he realized what had happened. He then took what he thought was a low dose. No one knew how potent LSD was, so this second dose turned out to be a significant dose at 0.25 mg (250 μg or 250,000 ng). At the time, no one ever heard of a "therapeutic" dose of any drug in the microgram range.
